Kiyotaka and Peko have never interacted with each other in canon, as they both come from two different entries in the series. While the two did attend Hope's Peak Academy around the same time, it is unknown if they had ever gotten the chance to meet or even know each other.
Personality-wise, Kiyotaka and Peko are very similar, as they both share many personality traits. They both have very serious and stern personalities, constantly worrying about the safety of their respective classes. They both even look serious, leading the protagonists of their respective games to be intimidated by their looks and personalities at first. However, they both also have a very soft side to them, seeing as how they are both very concerned with the safety of their classmates and despise the concept of the killing game, as they hate seeing their classmates get hurt. They both also have a common interest of having a certain classmate being very important to them and doing everything to keep them safe, even going to extreme measures to protect them. In Kiyotaka's case, it is Mondo, and in Peko's case, it is Fuyuhiko.
However, as similar as the two are, they are also very different. For one, in Chapter 2 of Danganronpa 2: Goodbye Despair, it is revealed that Peko is apart of the Yakuza, an infamous criminal organization. This is a conflict of interest with Kiyotaka, as Kiyotaka's talent is the Ultimate Moral Compass, so his morals may include being against criminal activity, especially those with the Yakuza. Also, in Danganronpa Another Episode: Ultra Despair Girls, it is revealed that Kiyotaka is related to the former prime minister of Japan, meaning he may have a lot of power in law, furthering his potential hatred for the Yakuza. However, these are merely theories.
The ship was mainly the result of both characters being featured together in a piece of promotional artwork for Danganronpa 1.2 Reload. The artwork featured Peko leaning in close to Kiyotaka, with the two of them wielding bamboo swords. The image alone was enough for some fans to start shipping the characters, as it made them realize the visual similarities between them. Most notably, their red eyes. Some other similarities include how both characters wear traditional Japanese school attire, and are known for their strong sense of morality. Meanwhile, some other fans came up with theories and head-canons about how the two may be related, and possibly separated at birth.
As one may expect, the ship isn't popular, as the characters have never met in canon. Nonetheless, there are still some who like to speculate about the possibility that the two may have some sort of connection in some way.
