In Vampire Ending, Reiji fights with his father. He kills him. Before Karlheinz dies, he gives his son his powers. Yui silently comforts him and promises to always be with him. Reiji is attacked by Carla and Shin but with his new powers he defeats them easily. Reiji and Yui live happily ever after, the only people remaining in the Demon World. Reiji promises to always protect her. Yui promises him to always suport him and stand by his side.
In Manservant Ending, Karlheinz overcomes Reiji in a fight. Richter appears to kill his brother who is saved by Reiji. Reiji passes away. Yui can't come to terms with it. She lives with the illusion of him, missing his touch. The illusion of him apologises but promises to be by her side till the end of time and after that. She promises him the same.
In Brute Ending, Karlheinz defeats Reiji in a fight. Reiji asks to be killed because he'd rather die than live with the uncertainty of what his father thinks of him. Karlheinz tell him he's useless if he can't become Adam and leaves. Reiji is heartbroken, feeling like a failure compared to Shu. In a fit of rage he denies ever loving Yui and stabs her. Looking at her dead body he apologises that he ever got involved with her. He maniacly laughs and then burst into tears.

In Vampire Ending, Reiji gains his father's powers. However, he finds it hard to control them. Yui and Reiji make a pact that she will kill him. Before she can do that, Reiji's brothers come, alongside Mukami brothers. They protect Yui and Reiji from Yui's father. They win. Some time later, the two live hapily ever after in their crowded manor.
In Manservant Ending, Yui kills Reiji with his permission. She gains Karlheinz's powers. She's attacked by Kino and her father but she survives. Using her powers, she restores Eden to its original form, in memory of Reiji.
In Brute Ending, when Yui is unable to kill Reiji, Kino attacks him. Reiji decides it can't end like this and decides to end the world. As everything goes up into flames, Reiji and Yui embrace each other. Yui is happy they're together, despite the physical pain she's experiencing.

Their relationship in the anime is very different to that in the games.
He's the 2nd person she meets in the Sakamaki household. He prevents Ayato from biting her. He asks her questions to try and understamd what is going on. When she learns he and his brothers are vampires, she tries to run away but fails.
The two rarely interact. He usually explains things to her or scolds her for her "lack of manners". He often stops Ayato, Laito and Kanato from biting her. He has no problem biting her himself, however. In one instance, he spikes her tea and proceeds to feed on her. As he does it, he thinks of his deceased mother, Beatrix.
Whilst in a trance induced by Cordelia, she has a vision of the brothers in their childhood. She sees kid Reiji who gets ignored by his mother in favour of his oldest brother, Shu.
When Cordelia awakens for the 1st time, he feels that something is off. When Cordelia awakens fully, she flirts with Reiji. She tells him he reminds her of Beatrix and that she deserved what happened to her. She kisses him while in Yui's body. When he calls her out on her lack of morals and her rotten nature, she slaps him in the face.
Together with his siblings, he tries to find way to save Yui's life. He makes a potion that succesfully brings her back to life.
